    Abstract:

    [Objective] Mutants of Isochrysis zhangjiangensis were screened for fast growth and high oil productivity. [Methods] Atmospheric and room temperature plasma jet mutation technique was used to mutate I. zhangjiangensis, and mutants were screened by different cultivation volume such as 96-well plates, flasks and bioreactors culture. [Results] After thirteen times passaged, mutant IM110020 was cultivated for 7 days in 600 mL bioreactor for evaluation. The maximum specific growth rate, cell density and lipid productivity could reach up to 0.72 d?1, 16 750×104 cells/mL and 109.8 mg/(L·d), respectively, which were 12.5%, 20.8% and 17.9% better than those of the wild strain correspondingly. [Conclusion] We obtained a mutant IM110020 with fast growth and high oil productivity, and the properties were stable after thirteen passages.
