[Objective] We aim to get a high-yield poly-β-hydroxybutyricacid (PHB) mutant strain. [Methods] We use composite multiple rounds of mutagenesis breeding of UV and diethyl sulfate and after primary and secondary screening from the original strain Pseudomonas koreensis PK3. [Results] It’s named Pseudomonas koreensis UVCN-18. After serially passed 9 times and fermentation 28 h, PHB production of the mutant was 15.94 g/L, Accounting for cell dry weight 69.54%. PHB production of the mutant was increased by 2.61 times compared with the original strain Pseudomonas koreensis PK3 (4.42 g/L), and the strain has a good genetic stability. [Conclusion] We use composite multiple rounds of mutagenesis breeding of UV and diethyl sulfate to get a high-yield PHB mutant strain.
