蓝细菌(Cyanobacteria)又称为蓝藻,属于蓝藻门(Cyanophyta),原核生物,无叶绿体及线粒体等细胞器的分化,根据形态表征、生长环境和代谢途径分类,大致有150属,2 000余种[1-2]。嗜热蓝细菌是一类分离于温泉地带,最佳生长温度为45 ℃或更高的蓝细菌[3],是能够在高温条件下生长的重要的初级生产者。生长在极端热环境中的微生物在能量代谢和物质循环中起着至关重要的作用[4]。蓝细菌在自然界中广泛分布,可生长在一些极端环境下,包括温泉[5]。这些自养原核生物在生物固碳、氧气生产和氮固定方面发挥重要作用[6],能够将光能和二氧化碳转化为高附加值产品,并且可以作为“自养型细胞工厂”合成生物燃料,被认为是非常具有研究潜力的微生物[7-11]。蓝细菌细胞中含有多糖、类胡萝卜素和藻蓝蛋白[7, 12-13]等多种高价值的生物活性物质,其中色素可以用作食品、化妆品和药品中的天然着色剂[13],蛋白质可以用作膳食补充剂,某些菌株甚至能够为人类和动物提供必需蛋白质[14-15]。近几年,蓝细菌藻蓝蛋白(Phycobiliproteins,PBPs)的研究引发学者关注,它是蓝细菌体内独特的捕光色素蛋白复合物,根据光谱特性分为3类:藻蓝蛋白(Phycocyanin,PC)、藻红蛋白(Phycoerythrin,PE)和别藻蓝蛋白(Allophycocyanin,APC)[16-17]。它可以作为一种天然着色剂[13];也可以利用其荧光特性应用于免疫学或作为实验研究中的荧光标记物[6];其强抗氧化性也使其在工业应用中受到重视[18-20]。从高温环境中分离和表征的蓝细菌具有更好的实际应用价值,具有较好的热稳定性[21-22]。此外,嗜热蓝细菌可为工业CO2利用提供强大的光合作用平台,在工业高温烟道气点源碳减排方面有可观的发展前景[23]。
虽然使用蓝细菌作为细胞工厂已变得越来越普遍,但研究仍使用标准的实验室模型生物而非潜在的生产菌株进行。纤发鞘丝蓝细菌属(Leptolyngbya)属于细鞘丝藻亚科(Leptolyngbyaceae)[24],迄今为止,Algaebase数据库显示Leptolyngbyaceae科中共有218种,其中Leptolyngbya有141种(http://www.algaebase.org/)。Kim等的研究发现Leptolyngbya sp. KIOST-1由于高效的生产力和高蛋白质含量,可用于生物质生产,并在营养方面有应用潜力[25-26]。有研究表明,Leptolyngbya菌株具有良好的固氮活性和合成生长素的能力,在制备微生物肥料方面有应用价值[27]。Leptolyngbya菌株可以有药物应用,生物活性化合物如Coibamide A、Dolastatin 12和Palmyrolide是用于治疗肺癌和乳腺癌的新型抗癌药物的重要成分[28]。Leptolyngbya菌株的分离和表征为未来研究和应用提供了更多的潜在可能。
蓝细菌在能源和环境应用方面显示出巨大的潜力,然而,蓝细菌资源仍然在很大程度上尚未开发。本项工作的目的是从温泉生态系统中分离鉴定蓝细菌菌株,表征2株目前研究较少的细鞘丝藻亚科(Leptolyngbyaceae)蓝细菌,评估其生物量生产力和细胞组成,验证是否是潜在的商业平台菌株[29]。这项研究为生物技术和环境应用中嗜热蓝细菌的分类和未来探索奠定了基础。
1 材料与方法 1.1 样品样品采自惠州地区横沥镇周围地热泉(N23°09′24.34",E114°36′54.40"),采用GPS地位。
1.2 培养基BG-11培养基(蒸馏水配制,g/L):NaNO3 1.5,K2HPO4 0.04,MgSO4·7H2O 0.075,CaCl2·2H2O 0.036,Na2CO3 0.02,柠檬酸0.006,柠檬酸铁0.006,EDTA-Na2 0.001,微量元素溶液1 mL。微量元素溶液(g/L):H3BO4 2.86,MnCl2·4H2O 1.81,ZnSO4 0.222,Na2MoO4 0.39,CuSO4·5H2O 0.079,Co(NO3)2·6H2O 0.049 4。
1.3 主要试剂和仪器氯仿、甲醇、苯酚、硫酸、D-葡萄糖、石油醚、丙酮、磷酸氢二钠、磷酸二氢钾,国药集团化学试剂有限公司;PCR相关试剂、基因组DNA提取试剂盒、DNA凝胶回收试剂盒及质粒小量提取试剂盒,上海捷瑞生物工程有限公司;大肠杆菌DH5α,天根生化科技(北京)公司;pMDTM19-T Vector Cloning Kit,TaKaRa公司;Whatman过滤膜、薄层色谱硅胶板,深圳市天翔化玻仪器公司;细菌蛋白提取试剂,北京康为世纪生物科技有限公司;Pierce™ BCA Protein Assay Kit,赛默飞世尔科技(中国)有限公司。C1000 TouchTM热循环仪,Bio-Rad公司;纳米光度计,Impeln公司;Epoch微孔板分光光度计,BioTech公司;超声波细胞破碎机,宁波科技生物技术有限公司;光学显微镜,OLYMPUS公司;便携式泵,圣斯特国际公司;马弗炉,上海阳光实验仪器有限公司;旋转蒸发仪,上海亚荣生化仪器厂。
1.4 温泉水样中嗜热蓝细菌的分离从惠州横沥镇周围的地热泉采集藻垫,转移到含有温泉水的50 mL无菌离心管。样品在12 h内转移到实验室进行分离。从地热泉中收集大约3 L水样,通过0.2 μm过滤膜进行浓缩后储存。将生物样转移到含有20 mL BG-11液体培养基[30]的培养皿中,在平均光照强度为7.1μmol/(m2·s)、光周期为16L:8D的45 ℃培养箱中进行富集培养。通过毛细管吸管洗涤和平板划线方法,分离纯化蓝细菌[23],纯化后的菌株每3周进行传代培养。
1.5 形态分析在100−1 000×放大倍数的显微镜下观察菌株的形态特征,主要观察以下特征:细胞尺寸,细胞排列成细丝或集落,细丝中的终端细胞形状,是否存在孢子、异形体或运动体等特定结构。根据形态特征鉴定蓝细菌[31-33]。
1.6 DNA提取、扩增和测序依据Singh等[34]的改良黄原酸盐法提取嗜热蓝细菌的基因组DNA,琼脂糖凝胶电泳检测基因组DNA质量。按即用型PCR试剂盒以蓝细菌基因组DNA为模板进行聚合酶链式反应(PCR)。引物8F (5′-AGAGTTTGATCCTGGCTCAG-3′)和1510R (5′-GGCTACCTTGTTACGA-3′)扩增其16S rRNA基因(约1 455 bp);藻蓝蛋白基因特异性引物为PhycoA_F (5′-ATGAAAACNCCKWTWACTGAAG C-3′)和PhycoA_R (5′-TTAGCTGAGGGCRTTRATG GCGTA-3′)扩增其PhycoA基因(约488 bp)。20 μL PCR反应体系:50 ng/μL DNA 1 μL,10 μmol/L上、下游引物各0.5 μL,High-Fidelity PCR Master Mix 10 μL和无菌去离子水8 μL。PCR反应条件:94 ℃ 3 min;95 ℃ 30 s,52 ℃ 30 s,72 ℃ 2 min,30个循环;72 ℃ 10 min。将PCR产物克隆到pMD19-T载体中,并转化到大肠杆菌DH5α中。阳性克隆送到深圳华大基因研究院进行商业测序以提供全长基因序列。16S rRNA基因序列提交到NCBI,登录号为MG822732、MG822734−44。
1.7 系统发育分析将测定的序列结果与GenBank数据库中搜集的蓝细菌参考菌株的16S rRNA基因和PhycoA序列,在MEGA 7软件中进行比对、编辑和修剪[35],绘制系统发育树。使用BioEdit 7 (http://www.mbio.ncsu.edu/bioedit/bioedit.html)对序列进行连接。用PhyML V3.0软件,采用最大似然法(ML)进行系统发育分析[36],其中使用的参数设置参考Tang等[37]。
1.8 生长条件和形态将筛选出的嗜热蓝细菌接种到含有350 mL BG-11培养基的500 mL锥形瓶中,45 ℃、100 r/min,光照为白光LED,70 μmol/(m2·s),光周期为16L:8D条件下培养。取对数生长期的嗜热菌株在光学显微镜下观察。培养20 d后,收集生物质用于随后的实验。
1.9 细胞成分分析 1.9.1 脂质取大约200 mg的冷冻干燥的生物质用于提取脂质[38]。加入氯仿:甲醇:水(2:4:1,体积比)混合物,重复提取生物质直至提取液无颜色。合并提取液后,倒入250 mL分液漏斗中,再加50 mL氯仿和50 mL水使之分层。收集氯仿层,用旋转蒸发仪浓缩获得脂质后称重。将水-甲醇层和萃取后的残渣收集,用于下一步糖分析。
1.9.2 糖类用苯酚-硫酸法,以D-葡萄糖为标准糖测定总糖含量,包括多糖、低聚糖和单糖组分[38]。将脂质提取后的残渣,用1 mol/L硫酸在100℃条件下水解2 h。水解产物稀释100倍用于之后的显色反应。亲水部分的含糖样品稀释10倍用于显色反应。显色反应是1 mL试样加入1 mL 6%苯酚溶液和5 mL 95.5%硫酸后,在490 nm处测量吸光度。
1.9.3 灰分将约80 mg生物质放入坩埚中,置于570±25 ℃的马弗炉中加热6 h。待坩埚冷却至约200 ℃后,转移至干燥器并冷却至室温。测量燃烧前、后的质量差来计算灰分含量。
1.9.4 蛋白质总蛋白用细菌蛋白提取试剂结合超声波细胞破碎机,输出9 s,占空比65%来提取。然后用BCA蛋白分析试剂盒进行定量分析。
1.9.5 色素将离心收集的生物质,加入100%丙酮匀化,进一步使用超声细胞破碎机破碎细胞2 min (输出9 s,占空比65%)后,置于4 ℃黑暗中保存20 min,4 ℃、12 000 r/min离心3 min后收集上清液,测定其在470、645、662 nm的吸光度。根据公式(1)计算色素含量[39]。

用薄层色谱法[TLC,石油醚:丙酮(7:3,体积比)溶剂系统和硅胶板]测定色素类型[40]。
用磷酸盐缓冲液(pH 6.8,含有Na2HPO4和KH2PO4)提取藻胆蛋白,采用冻融法结合超声破碎细胞[41-42]。通过分光光度法测量615 nm处的PC,652 nm处的APC和562 nm处的PE的吸光度,根据公式(2)计算藻胆蛋白含量[43]:

从惠州地区共分离出12株嗜热蓝细菌菌株,为了确定其分类学位置,对PCR扩增的16S rRNA基因和PhycoA序列经BLAST软件进行多序列比对,构建出系统发育树,如图 1−3所示。基于系统发育树,可以推断出其中6个菌株与Gloeocapsa sp. PCC7428的模型菌株非常紧密地聚类。因此,这些菌株很可能与先前文献报道的菌株相似。更为有趣的是,从惠州地区的温泉中分离出来的其他6株菌与目前研究较少的Leptolyngbyaceae的分支聚类[23],这使它们成为后续研究的重点。经过基于16S rRNA基因、PhycoA和16S rRNA基因+PhycoA基因序列的系统发育分析,我们发现这6株菌株表现出高度的系统发育同源性。因此,从中选择两株菌(PKUAC-GDTS1-24和PKUAC-GDTS1-29)进行更详细的生长特性表征,包括它们的形态特征和细胞组成。
2.2 形态表征
PKUAC-GDTS1-24和PKUAC-GDTS1-29菌株的形态特征相似,在液体培养基中,细胞排列成非常致密的簇,彼此附着形成毛状体或孤立的丝状物。毛状体为球形,表面有细小的、不明显的透明鞘,并且有少量假枝。在主细丝中细胞是等直径的或稍长于宽,而顶端细胞是圆形的。细胞呈蓝绿色,没有观察到异型细胞或运动细胞。这两种菌株的形态如图 4A、E所示,在液体培养基中嗜热蓝细菌形成小球。将蓝细菌切碎,放在显微镜下观察,细胞呈蓝绿色,形成厚垫(图 4B、F)或自由浮游细胞(图 4D、H)。
2.3 生物量及生长速率测定嗜热蓝细菌PKUAC-GDTS1-24和PKUAC- GDTS1-29的生物量及生长速率用培养20 d后的生物质干重计算。如表 1所示,PKUAC-GDTS1-24和PKUAC-GDTS1-29的生长速率分别为0.010 g/(L·d)和0.037 g/(L·d),总生物量产量分别为0.783 g/L和1.430 g/L。与表 1中其他类似菌株的生物质产量相当。

嗜热蓝细菌PKUAC-GDTS1-24和PKUAC- GDTS1-29的主要细胞组分(糖类、脂质、蛋白质、灰分、色素)组成非常相似,如表 1所示。PKUAC-GDTS1-24的脂质和蛋白质含量分别为21.40%和26.64%。糖类物质含量为36.42%,是其主要组成部分。PKUAC-GDTS1-29的脂质和蛋白质含量分别为23.92%和12.93%,糖类物质含量最高,为28.46%。2株嗜热蓝细菌的灰分含量相似,分别为24.20%和24.70%。
为进一步研究嗜热蓝细菌PKUAC-GDTS1-24和PKUAC-GDTS1-29,比较了其色素组成,如表 2所示。嗜热蓝细菌PKUAC-GDTS1-24和PKUAC-GDTS1-29中的色素组成分析显示两种菌株之间存在一些差异。PC是藻胆蛋白中的主要成分,在两个菌株中的含量为157.29 mg/g DW和374.86 mg/g DW。PE的含量最低,分别为23.79 mg/g DW和44.36 mg/g DW。APC的含量为118.99 mg/g DW和244.05 mg/g DW。类胡萝卜素是这些嗜热蓝细菌中的主要色素,在两种菌株中有较大差异,在PKUAC-GDTS1-24和PKUAC-GDTS1-29中含量分别为65.13 mg/g DW和18.87 mg/g DW。在本研究中,PKUAC-GDTS1-24的类胡萝卜素类型包括新黄素、紫黄素、胡萝卜素和叶黄素,PKUAC- GDTS1-29中有除叶黄素外的其他3种类胡萝卜素。

从惠州地区分离得到12株新的嗜热蓝细菌菌株,其中6株与Gloeocapsa sp.PCC7428模型菌株聚类,而另外6株与目前研究较少的Leptolyngbyaceae科的分支聚类。作为后续研究对象的2株嗜热蓝细菌PKUAC-GDTS1-24和PKUAC-GDTS1-29属于Leptolyngbyaceae,与被我们的研究小组描述为类Leptolyngbya属的第五簇菌株[23]的PKUAC-SCTA141、PKUAC-SCTA171和PKUAC-SCTA174的相似度最高(相似性为99%)。与从数据库中搜集的其他参考菌株相比,分离菌株与目前研究较少的Leptolyngbya sp. RV74 (相似性为96%)和Leptolyngbya sp. Greenland 7 (相似性为95%)相似性较高。而Leptolyngbya sp. RV74和Leptolyngbya sp. Greenland 7,除数据库中已知的16S rRNA基因外,缺乏系统的分类学研究,并且在系统发育树上常常与大量不可培养的蓝细菌聚在一起[23, 50]。尽管分离株在数据库中与属于Leptolyngbya属的一些菌株聚类,由于参考数据的质量不高,我们的分离株是否实际属于Leptolyngbya属存在不确定性。此外,根据Schleifer[51]的研究,16S rRNA基因序列相似性低于98.7%的原核生物可以被认为是不同的种。这些结果表明,在广东和四川的温泉中可能存在一种新型丝状轻度嗜热蓝细菌属或Leptolyngbya蓝细菌新种。Leptolyngbya菌株能够将光能和二氧化碳转化为高附加值产品,含有多糖、类胡萝卜素和藻蓝蛋白等多种高价值的生物活性物质,还可以有药物应用和用于制备微生物肥料,在未来应用中具有很大潜力[7, 25-28]。因此,本研究中确定的这些Leptolyngbya菌株为未来的工作研究提供了潜在可能。
已有的关于Leptolyngbya的研究主要集中在菌株的系统发育研究方面,对于其生理特性分析不足。本研究在分子鉴定基础上,选取类似Leptolyngbya的2株嗜热蓝细菌PKUAC-GDTS1-24和PKUAC-GDTS1-29进行进一步的生产力和细胞组分研究。就菌株生产力方面,PKUAC-GDTS1-29比PKUAC-GDTS1-24更有优势,2个菌株的生长速率分别为0.010 g/(L·d)和0.037 g/(L·d),与文献报道的其他Leptolyngbyaceae相似。Leptolyngbya sp. KIOST-1[26]的生长速率为0.024 g/(L·d),Leptolyngbya sp. ISTCY101[44]的生长速率为0.680 g/(L·d),Leptolyngbya foveolarum[45]的生长速率为0.085 g/(L·d)。与这些菌株相比,PKUAC-GDTS1-24和PKUAC-GDTS1-29的生长速率与Leptolyngbya sp. KIOST-1相近,比另外2种菌株慢。Kim等表示Leptolyngbya sp. KIOST-1具有高效的生产力,高蛋白质含量,可用于生物质生产[25-26]。因此,本研究的PKUAC-GDTS1-29菌株生产力更高,可以作为潜在的商业生产菌株。然而,由于这些研究中的生长条件包括光照强度、光周期、温度、培养基都不同(表 1),很难在没有更详细的生长优化研究情况下得出关于这些菌株生产力的绝对结论。
PKUAC-GDTS1-24和PKUAC-GDTS1-29的细胞组成中,糖类物质是其主要组分。根据之前有关菌株细胞成分的报道,我们比较了上述2株菌株与Leptolyngbya BL0902[46]、Leptolyngbya sp.KIOST-1[26]的细胞组成。Leptolyngbya BL0902含有38.8%蛋白质、14.6%糖类物质和5.8%脂质。Leptolyngbya sp. KIOST-1含有53.3%的蛋白质、24.8%的糖类物质和11.6%脂质。然而,我们的结果并不符合之前关于Leptolyngbya BL0902和Leptolyngbya sp. KIOST-1的报道,这2种菌株的主要成分是蛋白质,而PKUAC-GDTS1-24和PKUAC-GDTS1-29中糖类物质是主要成分,脂质含量更是高于Leptolyngbya BL0902或Leptolyngbya sp. KIOST-1。Leptolyngbya sp.菌株中灰分含量差异较大,Leptolyngbya BL0902中为10.3%,Leptolyngbya sp. KIOST-1中为37.6%。我们的菌株中灰分含量介于这2种菌株之间,分别为24.2%和24.7%。
当以总细胞蛋白质的百分比表示时,2种菌株中PBPs含量相似,PC是PBPs的主要成分。APC和PC的浓度超过总细胞蛋白的0.4%,其中PC含量分别为41.90 mg/g prot和48.46 mg/g prot,APC也是2种菌株的重要组分,分别为31.70 mg/g prot和31.55 mg/g prot。表 2的其他菌株中,PC和APC的含量显著低于我们的分离菌株,而PE的含量与分离菌株相似或更高。Leptolyngbya sp. KC45[47]的总藻胆蛋白含量为181.63 mg/g DW,其中PE是主要成分,含量为95 mg/g DW,APC和PC的含量分别为42 mg/g DW和44 mg/g DW。在Singh等[45]的研究中,Leptolyngbya foveolarum中PC、APC、PE的含量分别为42.5、30.0、28.5 mg/g DW。这些结果表明藻胆蛋白在Leptolyngbyaceae不同分支菌株中存在一定差异,但我们的分离株在藻胆蛋白生产积累方面具有明显优势,可以作为藻蓝蛋白生产的潜在菌株。
此外,我们对叶绿素和类胡萝卜素的含量进行了分析比较。在PKUAC-GDTS1-24和PKUAC- GDTS1-29中,类胡萝卜素含量分别为65.13 mg/g DW和18.87 mg/g DW,叶绿素a为0.97 mg/g DW和0.45 mg/g DW,叶绿素b为7.46 mg/g DW和2.15 mg/g DW。Joshi等[48]报道了Leptolyngbya sp.中的叶绿素a浓度为8.1 mg/g DW,类胡萝卜素浓度为4.5 mg/g DW。Trabelsi等[49]报道了Leptolyngbya sp.中类胡萝卜素含量为2.03±0.56 mg/g DW。还有研究表明Leptolyngbya foveolarum中的叶绿素a和类胡萝卜素含量分别为3.0 mg/g DW和5.2 mg/g DW[45]。嗜热蓝细菌PKUAC-GDTS1-24和PKUAC-GDTS1- 29中的叶绿素a含量明显低于表 2中其他菌株,但它们似乎合成了叶绿素b,而其他文献[45, 48]中的Leptolyngbyaceae菌株并没有此发现。PKUAC- GDTS1-24和PKUAC-GDTS1-29的类胡萝卜素的类型相似,但叶黄素仅在PKUAC-GDTS1-24中发现。
综上所述,基于16S rRNA基因、PhycoA和16S rRNA基因+PhycoA基因序列的系统发育分析表明,本研究中分离出了一组属于Leptolyngbyaceae科的轻度嗜热蓝细菌菌株,可能是一种新型丝状轻度嗜热蓝细菌属或Leptolyngbya蓝细菌新种。为阐明其与Leptolyngbyaceae其他成员的系统发育关系还需要更深入的分类学研究。关于此类菌株,目前的研究较少,仅有的一些系统发育研究不足以对它们的各项特性表征,本研究是第一次详细地研究此类菌株,并为进一步的生产研究奠定基础。PKUAC-GDTS1-24和PKUAC-GDTS1-29具有高效的生产力,并且能够生产比其他Leptolyngbyaceae菌株更高含量的具有价值的藻胆蛋白,可以作为潜在的生物质生产菌株,具有良好的研究和发展前景。而且它们生长形态呈现彼此附着的厚垫状,易于生物质收集,这对菌株的生产应用非常有利。
